ABSTRACT

Objectives : Gwanjul-bang(Guanjie-fang)-3 (GJB-3) is a prescription used clinically to treat rheumatoid arthritis. The purpose of this study is to investigate the immunity responses of GJB-3 to the human fibroblast-like synoviocytes isolated from patients with rheumatoid arthritis. Methods : Experimental groups were divided into 6 groups ; The hFLS were stimulated with hrTNF-α(100 ng/ml) and hrIL-1β(10 U/ml). Afterwards, They were treated with medium (CT), methotrexate (10㎍/ml, MTX) and GJB-3 (1, 10, 100㎍/ml) for 6 hours. The cells were harvested following the further incubation with [3H]-thymidine reagent for 4 hours. The WT group was cultured without cytokine stimulation. Results : 1. The proliferation of hFLSs was significantly reduced in 100㎍/ml dose. 2. The mRNA expression of TNF-α and the production of ROS was dose-dependently reduced in hFLS. 3. The mRNA expression of IL-1β, IL-6, IL-8, ICAM-1, NOS-Ⅱ, MMP-3 were reduced in high concentration. 4. The production of NO was significantly reduced in over 10㎍/ml dose. 5. The mRNA expression of TIMP-1 was dose-dependently increased. Conclusions : We found out that GJB-3 is considered to be effective in treating rheumatoid arthritis.
