Effects of Dipsaci Radix on Muscle Atrophy of MCAO Rats

ABSTRACT

Objectives : This study evaluated the effects of Dipsaci Radix on hindlimb muscle atrophy of stroke-model rats. Methods : Stroke-model was established in Sprague-Dawley rats using by the temporal occlusion of the middle cerebral artery(MCAO). Water-extract of Dipsaci Radix was administer orally, once a day, for 4 weeks. Then effects of Dipsaci Radix treatment were observed with changes of infarct size, neurological score, muscle fiber type and size, MyoD expression by immunohistochemistry and Western blotting. Results : 1. Dipsaci Radix treatment improved the aggravation of neurological score of MCAO rats, but it was not effective on infarct size induced by MCAO. 2. Dipsaci Radix treatment increased the ratio of muscle fiber type-I, MyoD expression and positive cells in soleus muscle of the contralateral hindlimb of MCAO rats. 3. Dipsaci Radix treatment was not effective on the muscle fiber size in soleus muscle of the contralateral hindlimb of MCAO rats. 4. Dipsaci Radix treatment reduced MHC-II expression in gastrocnemius muscle of the contralateral hindlimb of MCAO rats. Conclusions : These results suggest that Dipsaci Radix has a protective effect on muscle atrophy and motor disfunction of stroke patients.
