Objectives :This study was designed to investigate the effects of Lumbricus extract(LE) on the regional cerebral blood flow(rCBF) in ischemic rats, further to determine the mechanism of action of LE, and the effects that LE inhibits lactate dehydrogenase(LDH) activity in brain cells. Methods and materials :This study, ischemic rats were divided into total four group: control group(n=6), experimental group Ⅰ(LE treated group)(n=6), experimental group Ⅱ(LE treated group after pretreatment with indomethacin)(n=6), experimental group Ⅲ(LE treated group after pretreatment with methylene blue)(n=6). And the measurement that LE inhibits LDH activity in the damage to brain cells to N-methyl-D-aspartic acid(NMDA). The changes of rCBF were determinated by laser-doppler flowmetry(LDF), and LDH activity was determinated by microplate reader in vitro. Results :1. The rCBF was significantly improved by LE(10 ㎎/㎏, i.p.) during the period of cerebral reperfusion, compared with the control group. 2. The rCBF was significantly increased by LE after pretreatment with indomethacin(1 ㎎/㎏, i.p.), an inhibitor of cyclooxygenase, during the period of cerebral reperfusion, compared with the LE group, and rCBF was accelerated by LE after pretreatment with methylene blue(10 ㎍/㎏, i.p.), an inhibitor of guanylate cyclase during the period of cerebral reperfusion, compared with the control group. 3. LE significantly inhibited LDH activity in vitro in a dose-dependent manner. Conclusions :From the above results, these were suggested that Lumbricus had anti-ischemia action in connection with cyclooxygenase and might prevent the brain cells death through inhibited LDH activity.

Objectives :This study was to investigate the effects of Imyo-san treatment on the monosodium iodoacetate-induced osteoarthritis in rats. Methods :Arthritis was induced by injection of monosodium iodoacetate(MIA)(0.5 mg) into both knee joint cavities of rats. Arthritic rats were divided into control(n=8) and treated(n=8) group. Control group was taken distilled water and treated group was taken extracts of Imyo-san by orally for 20 days. At the end of experiment(20day after MIA injection), gross and histopathological examination on the articular structures of knee joints were performed. Blood cell counts and proteoglycan(PG) contents in articular cartilages were analysed. And also, tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) contents synovial fluids were measured by enzyme-liked immunosorbent assay(ELISA) method. Results :1. Body weight(g) of the treated group were increased significantly compared with control group at 15 and 20 days after injection. 2. Grossly, degree of osteoarthritis in the treated group was alleviated compared with the control group. 3. PG content in articular cartilage of the treated group was increased significantly compared with the control group. 4. Histopathologically, osteoarthritic scores of the treated group was decreased significantly compared with the control group. 5. TNF-α content in synovial fluid of the treated group was decreased significantly compared with the control group. Conclusions :On the basis of these results, we suggested that Imyo-san has inhibiting effects on the progression of arthritis in MIA-induced osteoarthritis model.

Objectives :The present study investigated anti-inflammatory effect of Artemisia Capillaris Thunberg in lipopolysaccharide-exposed rats. Methods :We divided lipopolysaccharide-exposed Sprague-Dawley rats into 4 groups. They were normal group, feed with 100 mg/kg Artemisia Capillaris Thunberg group, feed with 200 mg/kg Artemisia Capillaris Thunberg group and feed with 300 mg/kg Artemisia capilaris Thunberg group. They were administered for 6 weeks. We measured counts of red blood cell(RBC), the values of hemoglobin(Hb) and packed cell volume(PCV), plasma total protein concentration, albumin concentration, the ratio of albumin/globulin, the activities of plasma glutamic oxaloacetic transaminase(GOT), glutamic pyruvic transaminase(GPT), lactate dehydrogenase(LDH), the counts of white blood cell(WBC), the ratio of neutrophils, lymphocytes, monocytes, basophils, eosinophils, the concentration of plasma interleukin-1β(IL-1β), plasma interleukin-6(IL-6), plasma tumor necrosis factor-α(TNF-α), plasma interleukin-10(IL-10), the concentration of liver IL-1β and IL-6, TNF-α, IL-10. Results :Counts of RBC and the values of Hb and PCV, plasma total protein concentration and albumin concentration, the activities of plasma GOT, GPT and LDH showed no significant difference in the treatment groups. and the ratio of albumin/globulin was increased in Artemisia Capillaris Thunberg groups. The counts of WBC showed lower values in Artemisia Capillaris Thunberg groups than those of control group, In the ratio of neutrophils, Artemisia Capillaris Thunberg groups showed lower values than control group, however the ratio of lymphocytes was increased in the Artemisia Capillaris Thunberg groups. The ratio of monocytes, basophils and eosinophils were below 5%, and showed no characteristic trend. The concentration of plasma interleukin-1β(IL-1β), plasma interleukin-6(IL-6) and plasma tumor necrosis factor-α(TNF-α) showed a lower values in the Artemisia Capillaris Thunberg groups than those of control group, and the concentration of plasma interleukin-10(IL-10) showed no significant difference in the treatment groups. The concentration of liver IL-1β and IL-6 showed a lower values in the Artemisia Capillaris Thunberg groups than those of control group, however the concentration of liver TNF-α and IL-10 showed no significant difference in the treatment groups. Conclusions :The Artemisia Capillaris Thunberg groups gives positive results of anti-inflammatory response by lipopolysaccharide(LPS) derivation.
