AB - Objectives :
To investigate the neuro-protective effects of swimming in cold water in MCAo (Middle cerebral artery occlusion) rats.
Methods :
Rats were divided into six groups: SC (Sham plus control, N=7), SNS (Sham plus swim in normal temperature water, N=7), SCS (Sham plus swim in cold water, N=7), MC (MCAo plus control, N=7), MNS (MCAo plus swim in normal temperature water, N=7), MSC (MCAo swim in cold water, N=7). The MCAo rat model (permanent focal cerebral ischemia) was produced by a modified intra-luminal suture method as. From 4d after MCAo induction, SNS, SCS, MNS, MSC rats were participated in regular swim exercise during 30min. To measure the infarct volume, TTC staining were used and western bolt were conducted to determine COX-2 protein of expressions. To investigate level of behavioral motor function, all rats were completed Limb Placing Test and Balance Beam Test at before and after experimental exercise.
Results :
The infarct volume were significantly lowest in MSC among MCAo operated rats. COX-2 protein of expressions in MSC showed the lowest level significantly. There were significantly positive correlation between COX-2 expressions and infarct volumes. In Limb Placing Test and Balance Beam Test, behavioral motor functions were significantly decreased by MCAo induction and improved in regular swim exercise group.
Conclusions :
The present study demonstrates the effect of cold swimming training in improvement of behavioral motor function by reducing infarct volume and decreasing expression of COX-2 after ischemia.
