Objectives : This study was carried out to investigate the effects of Bee-venom treatment on the expression of NOS, c-fos, serotonin and substance P in central nerve system of monosodium iodoacetate(MIA)-induced osteoarthritic pain model. Methods : Arthritis was induced by injection of MIA(0.5mg) into knee joint cavities of rats. Arthritic rats were divided into control(n=8) and treated(n=8) group. Control group was injected with normal saline once a day for 20 days, while treated group was injected with Bee-venom extract once a day for same duration. Injected point of Bee-venom and saline was subcutaneous tissue at Tokpi(ST35) acupoint. Histochemical study of NADPH-d and immunohistochemical study of c-fos, serotonin and substance P in brain and spinal cord were performed at 20 days after MIA injection. Results : 1. The numbers of NADPH-d positive cells in superficial dorsal horn of spinal cord of treated group(24±4) was significantly (p<0.01) decreased compared with control(33±5). The numbers of NADPH-d positive cell in dorsolateral periaqueductal gray matter of treated group(114±18) was significantly(p<0.01) decreased compared with control(143±14). 2. The numbers of c-fos positive cells in dorsal periaqueductal gray matter of treated group(56±8) was significantly(p<0.01) decreased compared with control(78±13). The numbers of c-fos positive cells in paraventricular thalamic nucleus of treated group(64±14) was significantly decreased compared with control(88±27). 3. The numbers of serotonin positive cells in median raphe nucleus of treated group(104±15) was significantly(p<0.05) decreased compared with control(139±34). Conclusions : On the basis of these results, we concluded that Bee-venom treatment has inhibiting effects on the pain transmission in monosodium iodoacetate-induced osteoarthritic pain model in rat.

Objectives : In recent years, most research has focused on anti-aging and the extension of human life. The prevalence of obesity at all ages is becoming an epidemic around the world. Recent research has shown a positive effect on anti-aging, obesity. Methods : To identify the antioxidant effect, there were four independent experiments were carried out. They were DPPH radical scavenging activity, Nitric oxide(NO) radical activity, Superoxide anion radical scavenging activity and using Bovine serum albumin(BSA). One independent experiment was conducted to identify the candidate gene responsible for the gene expression profile of 3T3-L1 preadipocytes by Oligonucleotide chip microarray. Results and Conclusions : 1. The results showed that the ability of DPPH radical's elimination was increased by the rising concentration of Mori Fructus. When the concentration of Mori Fructus was 2.5 mg/ml, the ability of the DPPH radical's elimination was at a maximum. 2. The ability of the NO radical's elimination was maximum when the concentration of Mori Fructus was 5 mg/ml. 3. The ability of Superoxide anion radical's elimination was maximum when the concentration of Mori Fructus was 0.3125 mg/ml. 4. Antioxidative effects with BSA increased by the rising concentration of Mori Fructus. 5. Eleven kinds of gene(C3, Npy1r, Sort1, Agrp, Mod1, Cebpa, Hmox1, Hspb2, G6pd2, Tgfb1, Cebpb) expression increased and two(Ghr, Pik3r1) decreased when Mori Fructus extract was added. So I guess that Mori Fructus has effects of antioxidation, anti-obesity, etc.

Objectives : To investigate the neuro-protective effects of swimming in cold water in MCAo (Middle cerebral artery occlusion) rats. Methods : Rats were divided into six groups: SC (Sham plus control, N=7), SNS (Sham plus swim in normal temperature water, N=7), SCS (Sham plus swim in cold water, N=7), MC (MCAo plus control, N=7), MNS (MCAo plus swim in normal temperature water, N=7), MSC (MCAo swim in cold water, N=7). The MCAo rat model (permanent focal cerebral ischemia) was produced by a modified intra-luminal suture method as. From 4d after MCAo induction, SNS, SCS, MNS, MSC rats were participated in regular swim exercise during 30min. To measure the infarct volume, TTC staining were used and western bolt were conducted to determine COX-2 protein of expressions. To investigate level of behavioral motor function, all rats were completed Limb Placing Test and Balance Beam Test at before and after experimental exercise. Results : The infarct volume were significantly lowest in MSC among MCAo operated rats. COX-2 protein of expressions in MSC showed the lowest level significantly. There were significantly positive correlation between COX-2 expressions and infarct volumes. In Limb Placing Test and Balance Beam Test, behavioral motor functions were significantly decreased by MCAo induction and improved in regular swim exercise group. Conclusions : The present study demonstrates the effect of cold swimming training in improvement of behavioral motor function by reducing infarct volume and decreasing expression of COX-2 after ischemia.
