ABSTRACT

Objectives : The aim of this study was to investigate the apoptotic effects and mechanisms of Naetakcheongeum-san(Neituoqianjin-san) extract (NCSE)-treated on HL-60 leukemic cells. Methods : It was observed that NCSE induced apoptotic changes in a dose-dependent patterns, which was confirmed by DNA fragmentation and an increase of annexin V+/PI- stained cell population. It was monitored that the activation of caspase-3, -8, -9 enzyme activities and the cleavages of those in NCSE-treated HL-60 cells. We investigated the release of cytochrome c from the mitochondria to the cytosol and the bid cleavage in time- and dose-dependent patterns in HL-60 cells. To clarify the involvement of caspases, we used the inhibitors. Cells treated with the caspase-3 inhibitor, Ac-DEVD-CHO, and the caspase-8 inhibitor, z-IETD-FMK and the caspase-9 inhibitor, z-LEHD-FMK. Results and conclusions : It is suggested that NCSE-induced apoptosis is predominant through the activation of caspase-3 and -8 in HL-60 cells.
