ABSTRACT

This paper reviews recent advances and challenges in Drug Delivery Systems (DDS) as an essential strategy to enhance the therapeutic efficiency of conventional drugs. DDS aims to improve the stability, targeting ability, and controlled release profiles of drugs using various carriers such as polymeric nanoparticles, lipid nanoparticles (LNPs), and hydrogels. The paper discusses the classification of DDS by release mechanisms (immediate, sustained, stimuli-responsive), delivery routes (oral, transdermal, injection, mucosal, pulmonary, direct local delivery), and carrier types. It highlights practical applications in cancer therapy, chronic disease management, vaccine delivery, and treatment of neurological disorders. Representative case studies and experimental data demonstrate the superior therapeutic effects of DDS compared to free drugs. Finally, the paper addresses the current limitations of DDS, including in vivo stability and large-scale manufacturing, and proposes directions for overcoming these challenges through responsive polymers and advanced surface modification technologies.
